New Drug Daraxonrasib Doubles Survival Time for Pancreatic Cancer Patients
For decades, pancreatic cancer has held the title as one of the world’s deadliest cancers with very limited treatment options. Data from 2015 to 2021 indicates that around 97 per cent of patients diagnosed with metastatic (advanced-stage) pancreatic cancer died within five years. This cancer is exceptionally lethal due to the lack of effective screening tests and because it rarely causes symptoms in the early stages. However, this bleak treatment landscape is now beginning to change dramatically with the arrival of a new drug called daraxonrasib. In a Phase 3 clinical trial involving 500 metastatic pancreatic cancer patients, daraxonrasib proved capable of doubling patient survival time and slashing the risk of death by up to 60 per cent compared to standard chemotherapy methods. Genetically, over 90 per cent of pancreatic tumours are triggered by a mutation in a gene called KRAS. This gene functions as a switch that regulates cell growth. When mutated, the switch remains permanently active, instructing cancer cells to divide uncontrollably. For decades, scientists considered the structure of the KRAS protein impossible to target with drugs, calling it ‘undruggable’ because of its extremely smooth surface. Consequently, treatment has relied solely on heavy chemotherapy which damages healthy cells and causes severe side effects. Daraxonrasib introduces a new, precision approach. This pill, taken daily, works by binding to the cyclophilin A molecule inside the cell. This complex bond is then able to lock onto the active KRAS protein and disable its ability to send signals for cancer cell division. The clinical trial data was presented by the developing company, Revolution Medicines, on May 31, 2026. The results showed that daraxonrasib succeeded in extending the median patient survival time from 6.7 months (with chemotherapy) to 13.2 months after diagnosis. Although the drug causes common side effects such as skin rashes (experienced by over 86 per cent of patients), mouth sores, diarrhoea, and nausea, patients reportedly experienced a better quality of life with reduced pain. Patients also tended to persist with this drug treatment better than with conventional chemotherapy. Following the publication of this official data, Revolution Medicines will immediately apply for formal approval from the United States Food and Drug Administration (FDA) and other global regulatory bodies. Given that pancreatic cancer is extremely difficult to treat, breakthrough therapies like daraxonrasib typically receive special priority review so they can be used in health clinics within the coming months.
